Glutathione (GSH) is the major endogenous antioxidant produced by the cells. In ADMET studies, the potential toxicity of a drug candidate can be evaluated by testing the reactive intermediate formation with GSH. A representative reaction involves in vitro incubation of a drug candidate with an excess of GSH in hepatic microsomes or hepatocytes.
For structural analysis such as NMR, the purification and isolation of a sufficient amount of GSH-conjugates can be successfully obtained with AFFINIMIP® SPE Prep GSH Adducts.

Results : Total-ion-current profile obtained after an extraction on AFFINIMIP® SPE Prep GSH Adducts 10g. Purification of an incubation of 10mL containing 5mM of GSH.

GSH Adducts: No GSH and drug candidate observed
